Objectives: The aim of this study was to assess the association between dental fluorosis and the COL1A2 gene polymorphism among a sample of a Tunisian population. Methods: It is a case-control study established among the Tunisian population. Two groups were involved. The Control group included patients without dental fluorosis. The case group included Patients with dental Fluorosis. For each participant of both groups, 3 ml of Venous blood was collected. The DNA was extracted following the salting-out method. It was, then, was genotyped using Polymerase Chain Reaction-Restriction fragment length polymorphism (RFLP-PCR). Results: A total of 95 patients were included in the survey for both groups. The control group included 44 participants with 23 males and 21 females. Their mean age was 33 ±10 years. The case group included 51 participants with 23 males and 21 females. The prevalence of C/C genotype was 22.7% among the control group. The prevalence of A/C genotype was 31.8% and the prevalence of A/A genotype was 45.5%. Among the case group, the prevalence of C/C genotype was 39.2%. The OR was 1.17 for the AC genotype. It was 5.71 A/A genotype. The difference of their distribution was statistically between the case and the control groups. Conclusions: The current investigation demonstrated a considerably significant association of COL1A2 ( rs 412777) polymorphism in the COL1A2 gene with dental fluorosis among the Tunisian population.
