Acemannan Increased IL-6 and IL-8 Secretions in Primary Gingival Fibroblast

Objective: To investigate effect of acemannan on IL-6 and IL-8 expressions in primary human gingival fibroblasts (HGFs). Method: HGFs (6x104cells/well) were grown to confluence in 24-well tissue culture plate.  Cells were changed to serum-free medium for 3 h, two times.  After that, the cells were treated with acemannan (0-10 mg/ml) for 24 h. Control cultures received the same volume of serum-free DMEM only.  At 24 h of incubation, culture supernatant was collected. The levels of IL-6 and IL-8 secretions were measured using ELISA kits. The three independent experiments were performed. The data were presented as Mean ± S.D. and analyzed by using one-way ANOVA (SPSS program; p-value ≤ 0.05). Result:  Treatment with acemannan (10 mg/ml) significantly induced IL-6 and IL-8 secretions (p < 0.05).  Acemannan increased IL-6 and IL-8 expression by 3.6 and 6.8 folds, compared with non-treated group, respectively. Conclusion:  Acemannan induced IL-6 and IL-8 synthesis in primary human gingival fibroblasts.
