Effect of  Thai Medicinal Plant Extracts on Oral Cancer Cellline

Objectives:

The aim of this study is to determine the effect of these plant extracts on human squamous carcinoma cell line (HSC4)

Methods:

The cytotoxicity and anti- proliferation effect of water extracts of Zingiber officinale Roscoe, Albizzin myriophyll, Benth, Nigella sativa Linn., Mesua ferrea  L. and Lonicera japonica Thunb. were tested on human squamous carcinoma cell line (HSC4) and human gingival fibroblasts (ScienCelTM) using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method.  Then, a non-cytotoxic dose was chosen for each plant extract and used to determine anti- migration activity by scratch assay.For the scratch assay, cells were incubated with each extract in DMEM at the concentration of 10 mg/ml then incubated for 24 h.  After incubation, cells were stained with toluidine blue O. Area of cell migrated into scratch area were determined using ImagePro Plus Program. Migrating distances of leading front of cells were measured.

Results:

The results showed that the IC50 of Zingiber officinale Roscoe, Albizzin myriophyll Benth, Nigella sativa Linn., Mesua ferrea  L. and Lonicera japonica Thunb. for HSC4 cell line were  9 mg/ml, 51 mg/ml, 34 mg/ml, 16 mg/ml and 15 mg/ml  respectively.  The water extract of Zingiber officinale Roscoe, Albizzin myriophyll, and  Mesua ferrea  L. at the concentration of 10 mg/ml was found to possess anti- proliferation activity.  All extracts could inhibit oral cancer cell migration.

Conclusions:

Zingiber officinale Roscoe extract was found to possess significant anti-cancer activity and all extracts may be good candidates for further drug development for treatment of oral cancer.
