Bacterial-Plaque and Cytokine of PICF on Dental Implant

The clinical examination and radiography can not be used as a marker of early failure of dental implant. Objectives: To investigate the potency of Colony Forming Unit (CFU) of black pigmented bacteria number on dental plaque and the concentration of Interleukin 1 beta (Il-1β) on Peri Implant Crevicular Fluid as an early marker of dental implants condition to anticipate the prognosis. Materials and Methods: Before being enrolled in the study, respondents had approved an informed consent. Twenty seven respondents with healthy dental implant were participated as the subjects on the clinical trial. Dental implant plaque samples were anaerobically incubated on Brucella blood agar, 37oC, 3 X 24 hour, before the Black pigmented bacterial colonies were counted.  Black pigmented colonies were identified  using Polymerase Chain Reaction (PCR) method. Twenty-seven samples of Peri Implant Crevicular Fluid (PCIF) was tested for the amount of total protein using Bradford methods. Concentrations of Il-1β was checked using ELISA kits (Koma Biotech Inc). Correlation between the Il-8 expression on the peri-implant crevicular fluid with the CFU of black pigmented bacteria of dental implant plaque were analysed using Pearson correlation. Results: From the 27 samples, the CFU of Black pigmented bacterial were 8-258 and identified as Porphyromonas gingivalis. The concentration of total protein was 0.689 – 360.495 µg/ml, the concentration of Il-1β was 0 – 88.24 pg/ml. Conclusions: The CFU of black pigmented bacteria of peri-implant plaque is significantly correlated to Il-1β concentration in peri-implant crevicular fluid. Both can be exist as candidate markers of dental implants condition.
