Acemannan induces cementoblast proliferation, alkaline phosphatase activity and collagen expressions

Objective: To investigate effect of acemannan on proliferation, alkaline phosphatase activity, and type I collagen expression in cemetoblast cell line. Material and Methods: Cementoblasts were grown in 10% FBS, alpha-MEM supplemented with 10 ug/ml ascorbic acid, 0.1 uM beta-glycerophosphate. Cells were treated with various concentrations of acemannan. The [3H] incorporation assay, biochemical assay, and ELISA were used to determine new DNA synthesis, alkaline phosphatase activity and type I collagen expression, respectively. The data was presented as mean and standard error. The results were analyzed by ANOVA. Values of p<0.05 were considered as statistically significant. Results: Acemannan significantly increased cementoblast proliferation, alkaline phosphatase activity, and type I collagen secretions, compared with the untreated group. Conclusion: From our data suggest that acemannan could stimulate cementoblast proliferation, differentiation, and extracellular matrix formation.
