Histomorphometric Tissue Response Study of Hybrid-NanoForsterite/Poly Lactic Acid Scaffold (40 vs. 70% Porosity) in Rats
Objectives: Poly lactic acid (PLA) is a conventional thermoplastic polymer applied to develop 3D scaffolds. We designed the 3D scaffold of PLA microstruts coated it with nanocomposite gelatin forsterite fibrous layers with 40% and 70% porosities. We hypothesized that this might improve the mechanical strength and biological performance. Methods: In this animal clinical study, 36 round defects (r=4mm) were created in the skull of rats and rates were randomly distributed into either 40% (G1),70%( G2) or no scaffold groups (G3) in 1:1:1 ratio. The rats were sacrificed after 4 or 8 weeks and obtained specimens were assessed for histological and histomorphometric comparisons. Results: The mean percentage of total bone formation were 33.58 ± 4.32, 37.67 ± 6.20, and 33.67 ± 2.30 at 4 weeks and 34.25 ± 1.94, 37.33 ± 4.30, and 35.17 ± 4.139 at 8weeks P in G1, G2,G3. Respective groups showed statistically insignificant difference in degrees of inflammation of 1.83± 0.29, 2.16± 0.51, 1.92± 0.66 at 4 weeks and 2.16± 0.25, 2.25 ±0.88 and 2± 0.31 at 8 weeks (P>0.05). Angiogenesis at 4 weeks and 8 weeks were as the following for G1,2,3 groups: 4.58 ±1.43,4.83± 1.63, 5.33 ±1.83 and 7.84 ±2.46, 4.17± 1.29, 5.50± 1.48. Except for angiogenesis in 8wks (P=0.011), all other comparisons were not statistically significant (P>0.05). Conclusions: The findings revealed that the designed scaffold is biocompatible. 70% porosity were generally superior to 40% in terms of total bone formation. Major bone formation happened during the first 4 weeks.
