The Expression of Some Cytokine on Osteoblast Cell Seeded in Nano Chitosan-Carbonate Apatite Scaffold
Objectives: To observe the enchancement of some cytokine expression on osteoblast cells which is seeded in nano-chitosan scaffold as bone regeneration.
Methods: The increased expression of BMP-2, FGF-2, ALP, TGF-β1 performed by immunohistochemistry method that has been previously done seeding on osteoblast cells 2 x 106 in nano-chitosan scaffold, thereafter, incubating with autoclave 37°C for 3, 5 and 7 days. Paraffin blocks were carried out on the sample and observe the paraffin block on day 3, 5 and 7.
Results: The results of data analysis using Anova test results obtained by p < 0.050. This shows there is a difference between the significance of day 3, 5, and 7.
Conclusions: An increase in the expression of BMP-2, FGF-2, ALP in the osteoblast cells seeded in nano chitosan-carbonate apatite scaffold on day 3, 5 and 7. While increased expression of TGF-β1 occurred on day 3 until day 5, the day 5 until day 7 was relatively stable. The combination of nano chitosan-carbonate apatite scaffold structure and have good retention for osteoblast cells proliferation.
