SOD and CAT Enzymatic Regulation of Odontoblast Pulp Cells
Objectives: This research objective was to prove the enzymatic regulation of superoxide dismutase (SOD) and catalase (CAT) enzyme to maintain the homeostasis of the pulp cells. Methods: Using the pulp tooth of six-months-old Sprague Dowley mice (n=42; Male; 300-350gr), systemically anesthetized (Ketamine-HCl:diazepam; 100mg:10mg; 0,22cc/kg) for the experiment unit. Those mice were divided into two different groups, control group and treatment groups, both prepared with class I cavity and filled (GIC - FUJI IX). The treatment groups were exposed with resin 2-hydroxyethyl dimethacrylate (HEMA, 97% - Zigma Aldrich) at 0,016µg/ml for three periods of time (24, 48 and 72 hours). Decalcified with EDTA (10%, 60 days, 293K). Measured by immunohistochemistry protocol using antibody anti SOD and CAT (400x/10). Results: The average number of cells which expressed both SOD and CAT in odontoblast pulp cells exposed by HEMA solutions were increased rather than control groups. The mean and deviation standard result for expression of SOD periodically at 24, 48, and 72 hours were 5,000±1,290; 5,4286±0,975; and 5,4286±1,812. Whereas, for CAT the result was 5,7143±1,2535; 6,2857±1,2535; and 6,8571±1,5735. Kolmogrov Smirnov test showed normal distributed value (p>0,05). Levene’s test manifested a homogeneity (p>0,05; p=0,200). Multivariate statistical analysis test (ANOVA-Tukey HSD) showed the increasing in SOD and CAT expressions both in each group treatment and control groups (p>0,05). Conclusions: The exposure of HEMA 0,016µg/ml upregulated the expression of both SOD and CAT enzyme for 24, 48, and 72 hours. It means that HEMA could induce innate immunity in odontoblast pulp cell.
