Effect of Daun Ungu (Graptophyllum pictum (L.) Griff.) Extract Gel on The Number of Macrophages on Periodontitis Wistar Rats (Rattus norvegicus)

Objectives: To investigate the effect of Daun Ungu (Graptophyllum Pictum (L.) Griff.) ethanol-extract gel treatment as an anti-inflammatory agent on the number of macrophages as an inflammatory indicator on gingival tissue of Wistar rats (Rattus norvegicus) with periodontitis.
Methods: Twenty-four male Wistar rats were randomly divided into 5 groups: Control Group (n=4), Negative-Control Group (n=4), and 3 Treatment Groups (n=6 each). The periodontitis was induced by 0.2 ml Aggregatibacter actinomycetemcomitans 109 CFU on the right lower Molar gingival sulcus three times for a week. The Negative Control group was only induced by A. actinomycetemcomitans, the 3 Treatment Groups were induced by A. actinomycetemcomitans and then received topically ethanol-extract gel of Daun Ungu (Graptophyllum pictum (L.) Griff.) on the gingival sulcus of periodontitis region daily for 3 days in 3 different concentrations: 7.5% (Treatment Group I), 15% (Treatment Group II), and 30% (Treatment Group III). The number of macrophages cells from gingival tissue were analyzed based on Hematoxcylin-Eosin (HE) staining for histopathologic examination. The data was analyzed using Kolmogorov Smirnov Test for the normality, Homogenity of Variances Test for homogenity, and One-way ANOVA for the differences between groups.
Results: The number of macrophages cells were 1.50 + 0.577 (Control Group), 10.25 + 2.062 (Negative-Control Group), 5.00 + 2.530 (Treatment Group I), 3.76 + 1.211 (Treatment Group II), and 2.50 + 1.975 (Treatment Group III). This study showed a significant difference in the number of macrophages cells in each group (p < 0.05), except between Control Group with Tretment Group II (p=0.085), between Control Group withTreatment Group III (p = 0.414), between Treatment Group I with Treatment Group II (p = 0.228), and between Treatment Group II with Treatment Group III (p = 0.289).
Conclusions: Daun Ungu (Graptophyllum Pictum (L.) Griff.) ethanol-extract gel can decrease the number of gingival tissue macrophages in Wistar rats with periodontitis and the most effective concentration was 30%.
