Effects of Glucose Concentration on RANKL Expression in HPDL

Objectives: The epidemiological reports showed that diabetes patients have increased the alveolar bone resorption and attachment loss of periodontal organs. The present study was aimed to investigate the effects of glucose on the receptor activator of nuclear factor kappa-β ligand (RANKL) expression in human periodontal ligament (hPDL) cells.
Methods: HPDL cells were isolated from 3 independent healthy third molars with written informed consent for participation in this study. hPDL cells were treated with glucose at low (5.6 mM) or high (15, 25 and 100 mM) concentration. The cell number was investigated using MTT assay and mRNA expression of RANKL and osteoprotegerin (OPG) was determined by RT-PCR technique. Statistical differences were assessed using ANOVA at the significance level of 0.05.
Results: The results showed that 25 mM glucose has no effect on hPDL cell viability. High glucose significantly induced RANKL compared to low glucose, while not affect the OPG expression. The RANKL induced by glucose was attenuated when added inhibitor of glucose transporter (GLUT inhibitors) including cytochalasin B and phloretin.
Conclusions: These results suggest that role of glucose in bone remodeling process which may contribute to the prevention and treatment of periodontal disease associated with diabetes.
