Effect of Centrifugation Speed and Centrifugation Duration on Platelet Rich Plasma (PRP) Platelet Levels in Blood in Patients with Hypercholesterolemia

Objectives: to isolate, identify and analyze the effect of centrifugation rate and duration on blood platelet levels as an important growth factor for the success of periodontal surgery in patients with hypercholesterolemia
Methods: The study was conducted with pure laboratory experiments using several centrifugation methods in PRP making so as to obtain optimal platelet levels. In this study, there were 5 different methods of preparing PRP: Method 1: with two centrifugation, the first with 3000 rpm for 3 min 45 seconds, the second 3000 rpm for 13 minutes. Method 2: with twice centrifugation, the first 800 rpm for 15 minutes, the second 2000 rpm for 10 minutes. Method 3: with twice centrifugation, the first 1500 rpm for 10 minutes, the second 3000 rpm for 10 minutes. Method 4: with twice centrifugation, the first 2000 rpm for 7 minutes, the second being 2900 rpm for 10 minutes. Method 5: with one centrifugation of 3200 rpm for 15 minutes.Control: blood is precipitated without anti-coagulant to obtain blood serum and also assess the effect of platelet concentration in PRP on growth factor concentrations produced
Results: The highest growth factor concentration was obtained twice centrifugation, the first 800 rpm for 15 minutes, the second of 2000rpm for 10 minutes, with an increase in the TGF-β1 concentration of 4.36 times (436%) and increased concentration of PDGF-ββ by 4.40 times (440%) of the controls. While the most optimal method for getting the highest concentration of platelet is using two-stage centrifugation, first with a speed of 3000 rpm for 3 minutes and 45 seconds, second with a speed of 3000 rpm for 13 minutes. Platelet concentration was 6.09 times (609%)
Conclusions: There is the effect of the speed of centrifugation and duration of centrifugation to rombs platelet-rich plasma (PRP) thrombosis in blood in hypercholesterolemia patients
