Antibacterial Effect Between Calsium Hydroxide and Red Betel As Medicament
Objectives: To compare the antibacterial effect of calsium hydroxide and red betel as root canal medicament towards Enterococcus faecalis. In vitro study. Methods: Twenty premolar teeth were decoronized, then root canal was prepared and irrigated. The teeth were sterilized, and the root surface was layered with nail polish. Then Glucose Broth 40mL contained bacterial suspension were filled into the root canal and incubated. The germ samples were taken in 10mL, swapped on petri dish containing Glucose Broth Agar and germ colonial amount were calculates. The teeth were divided into 4 groups, the 1st group were given red betel paste, the 2nd group were given red betel and calcium hydroxide (Ca(OH)2), mixture, the 3rd group were given Chlorhexidine gluconate (CHX) 2% gel (positive control) and the 4th group were given Ca(OH)2 paste (negative control) . Those teeth were left untreated 7 days, then cleaned with ultrasonic and aquades, and rinsed with 5ml PBS. Total Plate count was used to determine the antibacterial effect. The result data were analyzed using one way anova and tukey test. The outcome of this study was total colony in one plate (CFU/mL). Results: The result showed significant differences between the treatment groups (p<0.05). The analysis using tukey test did not show significant differences between red betel paste (mean=7.87 ± 0.06 CFU/mL) with red betel Ca(OH)2 mixture (mean=7.92 ± 0.08 CFU/mL, p=0.823), and also between red betel paste with CHX 2% gel (mean=7.73 ± 0.14 CFU/mL, p=0.094). The red betel Ca(OH)2 mixture (mean=7.92 ± 0.08 CFU/mL) had significant differences with CHX 2% gel (mean=7.73 ± 0.14 CFU/mL, p=0.010). The comparison between red betel Ca(OH)2 mixture and Ca(OH)2 paste (mean=7.94 ± 0.05) also showed no significant differences (p=0.995). Conclusions: The effectiveness of red betel paste is almost equal to CHX 2% gel, which means it is more effective than red betel Ca(OH)2 mixture towards Enteroccoccus faecalis.
