Local and systemic evaluation of ligation as periodontitis induction in rat
Objectives: Periodontitis has an influence on the human systemic conditions. Several methods has been developed at this time to induce periodontitis in animal study. One of the method is by placement of silk ligature at lower incisiva cervical gingiva. The purpose of this study is to evaluate the local and systemic effect of ligature placement at lower incisiva to induce periodontitis. Methods: Forty male wistar rats were divided into two experimental groups. Periodontitis model groups were induced by placing silk ligature at cervical gingiva lower incisiva. The control group was only receiving injection of needle without ligation. Rats were sacrificed at 0, 1, 4, 7 and 14 days. Morphometric, bone radiograph, and hematological value changes were analyzed. Results: this study showed a reddish inflammation around the cervical gingiva and no pocket formation on the 1st day after ligation. Periodontal pocket start at day 4 after ligation. Periodontal pocket depth probe marked 2 mm, accompanied by bleeding. Measurements using a probe showed on day 7 after ligation showed 3 mm, and there is bleeding. The condition is getting worse on day 14. Radiograph showed that one day after the induction of periodontitis have not shown any changes in alveolar bone. There are radiolussen area in periapical alveolar bone in the lower incisor teeth bifurcation at day 4. Observation day 7 and 14 show the radiology area radiolussen wider area bifurcation lower incisor teeth. However, the results of the hematological analysis showed that neutrophil, eosinophil, basophil, lymphocytes and monocytes of peripheral blood of rat in the group and the sham periodontitis showed no significant difference (p> 0.05). Conclusions: this study showed periodontitis induction by using silk ligature has a local effect but no systemic effect.
