In-vitro Antibacterial Effects of Orthosiphon stamineus on Enterococcus faecalis

Objectives: To investigate the antibacterial effects of O.stamineus towards E.faecalis
Methods: Powdered whole O.stamineus plant was dissolved in distilled water and prepared in various concentrations. The antibacterial activity of O.stamineus was evaluated by measuring the minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and percentage of growth inhibition. Anti-adhesion activities of O.stamineus were determined by co-incubation with E.faecalis ATCC 29212 cultures in 96-well plates for 6 and 24h. Biofilm disruption activities were determined by adding O.stamineus extract into preformed E.faecalis biofilm grown for 48h. Positive (amoxicillin, 2.5% NaOCl) and negative (untreated E.faecalis culture) controls were used in this study. Anti-adhesion and biofilm disruption activities of O.stamineus were quantified using crystal violet staining. The effects on the morphology of E. faecalis grown as biofilm were assessed using scanning electron microscopy (SEM). T-test (p<0.05) was used to compare the mean absorbance values between O.stamineus treated samples and controls.
Results: O.stamineus showed a dose-dependent antibacterial action against E.faecalis, with the most effective inhibition (89.2%) at 12.5mg/mL. The MIC and MBC of O.stamineus extract were 25 and 0.78mg/mL, respectively. O.stamineus showed anti-adhesion activities at 3.13-0.78mg/mL following 24h of exposure. No biofilm disruption activity was observed. SEM analyses showed changes on the surface of E.faecalis cells in treated biofilm.
Conclusions: O.stamineus showed antibacterial effects against E.faecalis. However, the observed effect was more pronounced when E.faecalis was in suspension mode compared to biofilm mode.
