Inflammatory Regulation of Sticophus hermanii Extract on Porphyromonas gingivalis-Induced Periodontitis

Objectives: Periodontitis is a chronic inflammatory disease caused by microbial dental plaque. Porphyromonas gingivalis is one of major contributing pathogen associated with chronic periodontitis. Nuclear factor kappa B (NF-кB) is a member of the transcription factor family which plays a key role in coordinating the expression of genes in chronic inflammatory diseases, while interleukin-1β (IL-1β) is a potent inflammatory cytokine involved in many cellular functions, those are raised in periodontitis condition. Golden sea cucumber (Sticophus hermanii ) extract have been known to have antibacterial and anti-inflammatory properties regarding to its content. The aim of this study was to investigate the inflammatory regulation of Sticophus hermanii extract on Porphyromonas gingivalis-induced periodontitis by examine the expression of NFkB and IL-1β
Methods: The study was an experimental laboratories research with post test only control group design. Thirty male wistar rats aged 8-10 weeks were divided into 5 groups. Group 1 was normal group, while group 2-5 were periodontitis group induced by 109 Porphyromonas gingivalis ATCC 33277 three times on 4 days. Group 1 and 2 as control groups were given 0.2% CMC Na. Treatment group were given respectively Sticophus hermanii extract per oral 135mg/ kg BW once daily (group 3), applied topically on gingival sulcus 0.1ml of 3% extract gel (group 4) and 0.1ml of tetracyclin gel (group 5) once daily, in 14 consecutive days. The expression of NFkB and IL-1β expression were examined by immunohistochemistry. Data were analyzed by Manova and LSD test.
Results: The expression of NF-kB and IL-1β were raised in untreated P.gingivalis-induced periodontitis compare to normal group (p<0.05). Treatment with Sticophus hermanii extract peroral, topical and tetracyclin gel reduced the expression of NF-kB and IL-1β (p<0.05)
Conclusions: Sticophus hermanii extract regulates the inflammation process in P.gingivalis-induced periodontitis by decreasing the expression of NF-kB and IL-1β
