Strawberry Extract Effect towards Enterococcus-faecalis and Porphyromonas-gingivalis Biofilm in vitro

Objectives: To investigate the inhibition effect of strawberry extract towards the growth of monospecies and multispecies biofilm Enterococcus faecalis and Porphyromonas gingivalis in vitro.
Methods: The effect of strawberry extract on biofilm E. faecalis and P. gingivalis were observed with biofilm assay. E. faecalis ATCC 29212 and P. gingivalis ATCC 33277 were inoculated into BHI broth for 24h, in 37°C, and in anaerob condition. The suspension (24h old), were diluted to achieve the concentration of 107 cfu/mL (Optical dentisty 0.250-0.300). Further, 200 µL of E. faecalis and P. gingivalis suspension were distributed into micro plate wells, and were incubated for 24h to create biofilm. Strawberry extract with concentration of 100%, 50%, 25%, 12.5%, and 6.25% were added into well and the inhibition effect was observed after 1h, 3h, 6h and 24h. Biofilm without strawberry were used as negative control. Crystal violet and safranin (0.5%w/v) were used to count the biofilm mass. The remaining biofilm were counted using ELISA reader at 450nm and 490nm by adding 200mL of 90% ethanol.
Results: Strawberry extract inhibit the growth of monospecies and multispecies biofilm E. faecalis and P. gingivalis. The best concentration to inhibit each E. faecalis and P. gingivalis biofilm are 100% strawberry with 6h incubation (OD=0.860±0.028 and OD=0.187±0.029), otherwise on multispecies biofilm is 100% strawberry with 24h incubation (OD=0.138±0.014). One way Annova test result showed significant difference (p < 0.05) between several concentration and incubation time.
Conclusions: Strawberry extract is effective to inhibit E. faecalis and P. gingivalis biofilm as it can be one of the alternative therapy in preventing caries and periodontal disease. Future studies are needed to evaluate this effect.
