Supplementation of Sardinella longiceps Oil to Angiogenic Factors in Bone Grafting
Objectives: Sardinella longiceps oil is rich in omega-3 which act as immunomodulator and help the release of endothelial progenitor cells that produce angiogenic factors (VEGF and blood vessel) so that it could improve bone-defect treatment. The aim of study is to determine the effectiveness of supplementation Sardinella longiceps oil to angiogenic factors in bone grafting.
Methods: The experimental units (male wistar rat, n=20) were divided into two groups, control (K) and treatment (P) groups. Treatment groups were given Sardinella longiceps oil per oral seven days before the surgery to make the defect, and continued for seven days later. Control groups were not given Sardinella longiceps at all. All of the wistar rat were have two defects in their right femur, which is use hydroxyapatite (K+ and P2) and no use hydroxyapatite (K- and P1). Defect was made as big as half diameter of round bur in femur. Rats were sacrificed seven days after surgery, and femur were sliced transversally and performed immunohistochemistry staining for VEGF expressions examination and hematoxylin eosin stained for counting the amount of blood vessel lumen. Examination were done under light microscope. Data were analyzed using one-way Anova for blood vessels count and Kruskal-Wallis for VEGF expression.
Results: Sardinella longiceps oil increased the median/ modus of VEGF expression on P2 group : 3, compared to K- : 1 (p<0.05). It also increased the mean of blood vessel amount on P2 group : 21.29 ±11,73, compared to K- : 9,29±3,729 (p<0.05).
Conclusions: Supplementation of Sardinella longiceps oil was effective to increase angiogenic factors in bone grafting.
