Expression Analyses of BRAF, KRAS and PIK3CA Genes in Oral Leukoplakia
Objectives: Although oral leukoplakia (OL) is the most common oral potentially malignant disorders, its molecular biological properties are largely unknown. The aim of this study was to examine regulation frequencies of BRAF, KRAS, and PIK3CA genes by quantitative relative reverse transcriptase polymerase chain reaction (qRT-PCR) amplification in a cohort of OL specimens. These three genes were involved in kinase pathway for cell survival, and metastatis of abnormal cells and tissues. Methods: 25 OL samples and their corresponding match normal tissue (NT) specimens were obtained from histologically confirmed cases of OL. Regulation of BRAF, KRAS, and PIK3CA were evaluated by relative expression of fold compared with B-actin internal control gene of diseases and normal tissue specimens using Real Time PCR with respective gene specific primers. Results: The relative expression fold of BRAF, KRAS and PIK3CA were significantly upregulated in OL tissues (2.91, 1.92 and 2.48 respectively) than normal tissues. Whereas, PIK3CA was not significantly expressed compared with BRAF and KRAS genes in OL tissues. The mean ΔCt values for the BRAF among the OL tissues is 25.5±1.1, whereas NT is 28.58±1.4, KRAS for OL tissues is 28.5±1.2, whereas NT is 31.58±1.2, PIK3CA for OL tissues is 21.25±1.1, whereas NT is 36.58±2.1. Lesser Ct value indicates higher gene expression. Among OL samples, it was noticed that 50% revealed upregulation in gene expression pattern of BRAF and KRAS. Whereas, only 25% revealed potency of upregulation in expression of PIK3CA over NT. Conclusions: On comparison with NT, the expression of survival and metastatic genes were upregulated in OL, which implies, it has ability to grow faster and survive competent than NT. Expression of metastatic genes guides OL tissue to invade or invasive to other organs and tissue of oral regions. Expression of BRAF and KRAS plays significant role in development of cancer from OL.
