Tissue Response of TM-Joint to Arthritis Induction Methods in Rat
Objectives: Temporomandibular joint (TMJ) is a structure while involves in jaw movement. During inflammation, there are several inflammatory cytokines such as Interleukin 1-β (IL-1β) can (1) decrease ECM component production, (2) induce the expression of MMP-3 and (3) participate in osteoarthritis (OA) or rheumatoid arthritis (RA). Some of studies investigated the change of condyle after injected complete freund's adjuvant (CFA) which similar to OA. However, there are few studies to observe the effect of TMJ in collagen induce arthritis (CIA) which mimic RA. Even there were no studies to identify the TMJ tissue in disc, condyle and synovial tissue. Therefore, the aim of study is investigate the TM-joint response of CFA and CIA group in rat model. Methods: 12 Eight-week-old female Sprague-Dawley rats were divided randomly into 4 groups (n=3/group), injected CFA, CIA and normal saline as control at day 0. After sacrificed, TMJ tissues were stained by H&E staining and examined the ECM (Aggrecan, Collagen I and link protein), inflammatory (IL-1β and MMP3) gene expression in disc, condyle and synovial tissue by conventional PCR. β-actin as internal control. All data were presented as the mean±SEM. Using one-way ANOVA and post-hoc test (Dunnett's) to analyze the data. Results: In present study, we used alcian blue to confirm the injection position. In histological staining, the fibrocartilage tissue adapted to thickness in CFA group and became thinness in CIA group at day 7 compared to control group. In gene expression, there were significant difference in MMP3 of CFA and CIA disc (p=0.034; p<0.01), IL-1β of CFA and CIA synovial tissues (p=0.012; p=0.02). Inflammatory gene expression could not be detected in condyle. Conclusions: The fibrocartilage of condyle showed different thickness, especially in transitional and hypertrophic layers between CFA and CIA arthritis induction methods.
