Effectiveness Of Myostatin Knockdown On Jaw Muscle Activity

Objective: Myostatin, a member of TGF-β superfamily, is known as a negative regulator of cell growth and differentiation in skeletal muscle. RNA interference (RNAi) offers a novel therapeutic strategy by efficiently silencing target genes known to promote disease. This study evaluated the effectiveness of local administration of cationic liposome-delivered myostatin-targeting siRNA (Mst-siRNA-lipoplexes) on jaw muscle activity.

Methods: Mst-siRNA and scrambled (scr)-siRNA-lipoplexes were injected into the masseter muscles of dystrophin-deficient mdx mice, which model Duchenne muscular dystrophy. One week after injection, the masseter muscles were dissected for histometric analysis. To evaluate changes in masseter muscle activity, electromyographic (EMG) measurements were performed.

Results: Sectional area of masseter muscle fibers treated with Mst-siRNA-lipoplexes was significantly larger compared to the control injected with scr-siRNA-lipoplexes. The daily integral value of masseter muscle activity was significantly increased by knockdown of myostatin.

Conclusion: These data suggest that local administration of Mst-siRNA-lipoplexes may be a useful tool for treating muscle atrophy diseases including muscular dystrophy.
