Analysis of Cyclin D1 gene polymorphism in salivary gland tumors

Objectives: As a key regulator, Cyclin D1 plays a critical role in the transition from G1 to S phase of the cell cycle. We aimed to analysis the association between Cyclin D1 A870G polymorphism and increased susceptibility to salivary gland tumors in Chinese patients .

Materials and method: The patients include 4 cases of pleomorphic adenoma (including 2 men and 2 women), one case of adenolymphoma (man). Thirteen cases of non-tumor patient were used as control (including 4 men and 9 women). All patients were diagnosed at The First Affiliated Hospital of Dalian Medical University between 2008 and 2009. Genomic DNA from each case was isolated from oral mucosa cell with the GENMED Oral mucosa cells DNA Purification Kit (GENMED SCIENTIFICS INC. U.S.A). Cyclin D1 A870G polymorphism was analyzed by polymerase chain reaction. The 167 fragment spanning the A870G polymorphism in exon 4-intron 4 boundary was amplified with the primer. The sequence of forward primer was 5-GTGAAGTTCATTTCCAATCCGC-3, and the sequence of reverse primer was 5-GGGACATCACCCTCACTTAC-3 in intron 4. PCR products were sequenced to determine their genotypes.

Results: It was found that the genotype of 4 cases of pleomorphic adenoma and one case of adenolymphoma was AA (100%). In normal controls, 12 cases were AA genotype (92.3%) and 1 case (7.7%) was AG genotype.

Conclusion: Our data suggests that the Cyclin D1 A allele is associated with increased susceptibility to salivary gland tumors. Cyclin D1 may be an important target for the development of new strategies for cancer treatment and prevention.
