Antimicrobial Effect of Ursolic acid against Mutans Streptococci
Objectives: The purpose of this study was to determine the antimicrobial activity of ursolic acid against mutans streptococci isolated from oral cavity of Koreans. Methods: The antimicrobial activity was evaluated by minimal inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and time-kill curves on mutans streptococci. Results: MIC90 values of ursolic acid were 2 ug/ml for Streptococcusmutans, 4 ug/ml for S. sobrinus and 8 ug/ml for S. downei. MBC90 values of usrolic acid were 4 ug/ml against mutans streptococci. Time-kill curves were performed against S. mutans ATCC 25175T, S. sobrinus ATCC 33478T and S. downei. ATCC 33748T in the presence of ursolic acid at their respective MBC. Usrolic acid 2×MBC were bactericidal against S. mutans ATCC 25175T and S. downei. ATCC 33748T after 12 h of incubation whereas 4×MBC was bactericidal against S. sobrinus ATCC 33478T. We were also tested by MTT assay in human gingival fibroblasts (HGF) and periodontal ligament fibroblasts (PLG). Cytotoxicity values were 16 ug/ml on HGF and PGL for ursolic acid. Conclusions: The results indicate that ursolic acid have antimicrobial activity at mutans streptococci and can be useful in the development of oral hygiene products for the prevention of dental caries.
