Methods:Cytotoxicity,biocompatibility and cell secretary function were evaluated for the purpose of study the biologic properties. Firstly, human periodontal ligament cells(PDLCs) were separated and cultured on the modified collagen membrane. Cell growth rate was measured by MTT methods in order to quantitatively evaluate the cytotoxicity of the modified bionic collagen membrane. Secondly, for the purpose of biocompatibility of the modified bionic collagen membrane, adherences and spreading condition of PDLCs were observed by SEM. Thirdly, HYP and ALP and OC in supernatant of the culture mediums were measured by chromatometry and enzyme linked immunosorbent assay in order to evaluate cell secretary function.
Results: The toxicity gradation of the modified bionic collagen membrane in different concentration extract and different culture time were ranked from 0 to 1 grade. The biomaterial had no cytotoxicity to human PDLCs. There is no difference effect on the cell growth in higher concentration from lower concentrations (P>0.05). The human PDLCs adhered firmly on the surface of the modified bionic collagen membrane and proliferated rapidly. The adjacent cells were connected. The cells were accumulated growth. There was high concentration of HYP, ALP and OC in supernatant of the experiment group.
Conclusion: The modified bionic collagen membrane had no cytotoxicity to human PDLCs. Human PDLCs cultured on the loose layer of biomaterical membrane maintained its morphological and biological property. The biomaterial membrane may fulfill the security standards of medical biomaterials.
