The effect of SIgA on S.mutans biofilm

Objectives: It is proved that the targeted fusion DNA vaccine pGJA-P/VAX1 can stimulate the secretion of IgA(secretory IgA,S-IgA)in saliva and have effect of anti-caries. The purpose of this study is to detect whether the immunity effect of pGJA-P/VAX1 is that the SIgA antibody interfere the formation of S.mutans biofilm.

Methods: Study the kinetics of the antibody responses generated following targeted anti-caries DNA plasmid immunization .Use the SIgA to influence the biofilm cultivated by UA140:ƒ³(mutAp-rfp1),then use the CLSM to determine the depth of biofilm and CFU to determine the effect of SIgA on the adherence ratio of S.mutans.

Results:The result suggest that the the depth of biofilm of SIgA higher group and lower group has significant difference(P<0.05),the CFU have the same trend(P<0.05). SIgA produced by the targeted fusion DNA vaccine pGJA-P/VAX1 markedly effect the adherence of S.mutans and use this mechanism to have the effect of anti-caries , the binding of S.mutans and SIgA can be observed clearly under CLSM.

Conclusion: The data demonstrate that the SIgA produced by the targeted fusion DNA vaccine pGJA-P/VAX1 can prevent the adherence of S.mutans and have the effect of anti-caries by this way.
