Sensitive antibiotics of recombinant Streptococcus mutan JH1140 containing urease gene

Objectives:To investigate the sensitive antibiotics of the recombinant JH1140 which contains urease gene. Methods:The sensitive antibiotics of the recombinant Streptococcus mutan JH1140 were examed through sensitivity drug papers of eight common antibiotics chosen from the clinical application .The antibiotics include cefradine, clindamycin, penicillin, gentamicin, chloromycetin, ciprofloxacin, metronidazole, vancomycin. The recombinant Streptococcus mutan JH1140 was cultivated to 5°Á108/ml.Then the drug papers and the recombinant Streptococcus mutan JH1140( 50µl)were co-cultured in 37℃,90%N2,10%CO2 in the TSA plates for 24 hours. After cultivation ,the diameters of the central inhibition areas of the drug paper were measured .The means of the inhibition diameters were calculated for the same three samples. And the same method was used on the original Streptococcus mutan JH1140 which contains no urease gene.Compare the diameters of each antibiotic on the two bacterial by t-test. Results:The recombinant Streptococcus mutan JH1140 was sensitive to several common antibiotics, and the sensitivity effects against different antibiotics varied from each other. For each antibiotic, there was no significant difference between the recombinant Streptococcus mutan JH1140 and the original Streptococcus mutan JH1140 on the sensitive antibiotics(P>0.05). Conclusion: The recombinant Streptococcus mutan JH1140 were sensitive to several common antibiotics. And they could be controlled by sensitive antibiotics. The basic characteristics of recombinant Streptococcus mutan JH1140 can be used in further investigation of the application in replacement therapy.
