Objectives:
Enamel matrix derivative (EMD) represents a purified acid extract of pig enamel matrix and is widely applied in promoting periodontal or peri-implantitis tissue regeneration in recent clinic study. The molecules mechanisms within EMD stimulation surrounding oral implants are un-cleared. The purpose of this study is to investigate the influence of Enamel matrix derivative on proliferation, differentiation and cellular mediators in osteoblast cells on the implant surface
Methods:
This study examined the response of human osteosarcoma cell line MG63 cell to Enamel matrix derivative on the coarse-grit-blasted and acid-etched surface titanium implant disks (SLA). MG63 cells were seeded onto SLA implant surfaces with stimulate the different concentration of Emdogain. Proliferation was detected by cell number by the cell number kit-8. Alkaline phosphatase activity was measured colorimetrically using p-nitrophenyl phosphate as a substrate. Cellular cytokine production of OC and transforming growth factor beta-1 (TGF-â1) was detected in cell culture supernatants with immunoassays. Expression of the bone-associated genes osteoprotegerin (OPG) and Receptor activator of nuclear factor (NF-kB) ligand (RANKL) was determined by reverse transcriptionpolymerase chain reaction.
Results:
On the SLA implant surface, Emdogain inhibit the proliferation of MG63 cells, but then increase differentiation of MG63 cells. Emdogain enhanced the TGF-â1 production on the SLA implant surface. Emdogain enhance OPG expression but have no effect on RANKL expression by Real time RT-PCR.
Conclusion: Emdogain has been successfully used to effect the proliferation and differentiation of osteoblast on the surface of implant. Emdogain stimulates the production of TGF-â1 which further enhances production of the extracellular matrix. Emdogain enhance OPG but have no effect on RANKL expression which may result in physiological remodeling of bone tissue. So Emdogain can accelerate the regeneration of hard tissue in the peri-implantitis disease.
