Synthesis and in-vitro analysis of injectable bioactive glass hydrogel for periodontal bone regeneration

Objectives: As bioactive glass is known for osteoinduction activity, therefore, it can be used as an injectable to fill irregular periodontal bony defects. The aim is to characterize a novel injectable bioactive glass hydrogel.
Methods: A novel injectable bioactive glass hydrogel was synthesized via co-precipitation method. It was characterized physically and biologically to evaluate its phase purity, crystallinity and in vitro cytotoxicity. XRD technique has been used and for cytotoxicity evaluation, MTT assay was performed using Mouse pre-osteoblast cell line MC3T3-E1 and culture media with and without cells were used as positive and negative control respectively. The absorbance was measured at 590 nm. The samples were evaluated at day 1 and 7 in reference to positive, negative and control groups. Two-way analysis of variance was applied to do statistical analysis by using SPSS software.
Results: In XRD, a broad band was observed at 2θ ≈15°–39°. This band was a characteristic band for polymers. The absence of reflections peaks in XRD pattern, confirmed the presence of amorphous structure of bioactive glass powder. In MTT assay, the experimental material showed a continuous proliferation and cell growth. There was a statistically significant (p ≤ 0.05) increase of OD values of injectable hydrogel group related to positive control, and insignificant (p ≥ 0.05) OD values related to control bioactive glass group. The cells were viable till day 7. This indicated that injectable hydrogel has promoted the cell growth, facilitated proliferation and has no cytotoxic effect.
Conclusions: The prepared injectable bioactive glass had a non-crystalline and amorphous structure. It was not cytotoxic and completely biocompatible with the host tissue. It is anticipated that it will enhance the bone regeneration and will show promising effects for the hard tissue regeneration.
