Synthesis and in vitro Biocompatibility Evaluation of a Novel Bioactive Fiber Reinforced Dental Composite

Objectives: The aim of this study was to synthesize a novel biocompatible dental composite and to achieve this aim the objectives were to evaluate cell toxicity and monomer leaching and bacterial adhesion of this novel dental composite.
Methods: The novel dental composite reinforced with nano-hydroxyapatite (n-HA) and E-glass fibers was synthesized under inert environment. Four experimental resin-based composites with various concentrations of fillers (Exp-0, Exp-40, Exp-50, and Exp-60) were prepared and compared with commercial composite Filtek-Z350, 3M ESPE, Germany (CT-Z350) (in each group n=27). Biocompatibility was evaluated by in vitro cytotoxicity testing, by detection and quantification of the monomers release and bacterial adhesion testing. The cytotoxicity was assessed using MTT Assay by both direct (day 1, 3 and 7) and indirect methods (after 3 day incubation of extracts). The release of component monomers (in deionized water on day 1, 7 and 28) were evaluated by HPLC with methanol, water and acetonitrile gradient by PDA detector. Bacterial (S. mutan; ATCC-25175) adhesion was evaluated by CFU at 24h and 48h. The results were statistically analyzed by One way ANOVA and post hoc tukeys test for comparison.
Results: Both direct and indirect MTT assay evaluation demonstrated no cytotoxic effect on NIH/3T3 fibroblast cells and in parallel supported their viability in all experimental groups on each interval. The HPLC detected the release of monomers. Exp-50 showed highest monomer release among all study groups. All monomers showed manximum release at day 1. The number of active colonies grown on BHI agar were less for novel composite.
Conclusions: The experimental dental composite was found to be biocompatible and comparable with commercial material.
