Evaluation and Comparison of Antibacterial Properties Of Equia Coat Functionalized With Zinc Oxide And Titania Nanoparticles

Objectives: Recurrent caries was partly ascribed to lack of antibacterial properties in restorative materials and coatings. Titania and Zinc Oxide nanoparticles are considered to be broad-spectrum antibacterial agents. The aim of this study was to add antibacterial agents in commercial restorative coating material and to evaluate the antibacterial properties.
Methods: Total two hundred discs were prepared from EquiaTM coat containing 0%, 1% and 2% wt/wt nanoparticles (n=40). The antibacterial properties were analyzed by colony forming unit (CFU), crystal violet staining (CVS) procedures and Scanning Electron Microscopy (SEM). For CFU, diluted solutions of streptococcus mutans (ATCC 25175) and Lactobacillus acidophilus (ATCC 4356) were prepared. For this 0.01mL of each bacterial species was separately placed on discs. The liquid culture media was transferred to an incubator at 37oC for 24h, 48h and 72h. Thereafter, 0.01ml of solution from each tube was cultured on Brain Heart Infusion agar and MRS (De Man, Rogosa and Sharpe) agar plates and colonies were counted. To quantify the bacterial adherence the biofilm formed on discs was stained with crystal violet stain and absorbance was measured by a microplate reader. Data was analyzed by One way ANOVA and Post Hoc Tukey’s tests.
Results: EquiaTM coat discs containing nanoparticles exhibited significantly high (p<0.05) number of colonies as compared to control group (EquiaTM coat). The results of absorbance values were significant for both zinc oxide and titania nanoparticles discs (p<0.05) and these were later supported by SEM images.
Conclusions: These results suggested that EquiaTM coat with incorporated nanoparticles exhibited antibacterial properties for CVS behaved as antibacterial revolutionary coating but for CFU results are not very obvious and further studies are recommended to confirm its behavior.
