Acid resistance of human enamel treated with diode-laser and fluoride varnish: in-vitro atomic absorption spectrometry analysis

Objectives: To evaluate the efficacy of diode laser irradiation and fluoride varnish application on in vitro acid resistance of human tooth enamel.
Methods: An in vitro study was carried out on 40 human premolars to evaluate the enamel's acid resistance using an atomic absorption spectrometry analysis. A total of 40 enamel specimens occlusal surface were prepared from 40 human premolars and were randomly assigned to 4 groups: 1. Untreated (control); 2. 5% fluoride varnish application (Duraphat®); 3. High power Diode laser (Wiser doctor smile®) irradiation alone; 4. Diode laser irradiation + 5% fluoride varnish application. The specimens were then individually immersed in 5 mL of acetate buffer solution (0.1 mol/L, pH 4.5) and incubated at 37°C for 24 h, and the acid resistance was evaluated by determining the calcium ion concentration using an atomic absorption spectrometry. An ANOVA model was constructed (P-value <0.05), followed by Tukey's test for multiple pair-wise comparisons of mean values.
Results: Significant differences were found between the control group and the three groups (P < 0.001). Combining Diode laser irradiation with 5% fluoride varnish application had a better effect in decreasing the acid dissolution of calcium compared to fluoride varnish application or laser treatment alone.
Conclusions: The combination of high-power diode laser irradiation with fluoride in 5% varnish was more effective on the acid resistance of enamel, with which a higher degree of prevention could be achieved from the decrease in mineral dissolution.
