Endothelin 1 regulates Hey1 expression during mandibular patterning

Objectives: The mandibular development requires cell interactions between germ layers and cranial neural crest cells; interactions that are regulated by several signaling pathways. Within these pathways, Endothelin 1 (End1) and Notch signaling pathway, in particular the effector gene of Notch pathway -Hey1- has been involved in crucial cell events during organogenesis. The aim of this research was to determine the relationship between Hey1 and End1, during mandibular development.

Methods: Methods: Hy-Line W-36 chicken embryos were incubated at 38°C and staged according to the Hamburger-Hamilton (HH). The patterns expression of Hey1 and End1 in HH18 and HH21 stage were determined by in-situ hybridization, RT-PCR and immunofluorescence. Inhibition assays of Notch and End1 pathways were performed using DAPT and Phosphoramidon, respectively. All trials were performed in triplicate. All the experimental procedures that were carried out got the approbation by the ethics committee of the School of Dentistry of the National University of Colombia.

Results: Results: the transcripts of Hey1 and its protein are expressed in the dorsal and ventral domains of the mandibular prominence. The transcripts of End1 were detected in the dorsal, intermediate and ventral domains of the mandibular prominence. The inhibition assays of End1 pathway shown Hey1 ectopic expression in the intermediate domain of the mandibular prominence. The inhibition assays of the Notch pathway suggest that part of Hey1 expression could be mediated via the Notch pathway.

Conclusions: Conclusions: Hey1 can be an important factor in the regulation of facial formation, ensuring the dorsal and ventral identity of the mandibular prominence through its interaction with End1. The restriction of the Hey1 expression in the intermediate domain of the mandibular prominence may be due to a negative regulation by the End1 signaling.
