Effect of PDGF on proliferation of human PDL cells

Objectives: PDGF is a significant polypeptide mitogen for most cells of mesenchymal origin, we investigated which(or how) isoform of PDGF has the greatest mitogenic and proliferative biological activity on human PDL cells.

Methods: Human PDL primary cell culture, proliferative response measurement, DSP & DMP-1 Western Blot analysis

Results: 1. PDGF dimers increased proliferative response by PDL cells time- and dose-dependently, especially PDGF BB. 2. In DSP Western blotting, PDGF AA treatment group showed decreased levels of this protein compared with the control group. The expresson in the PDGF AB treatment group was similar to that in the PDGF BB group, and the expression in both of these groups was significantly higher than that in the control group. 3. In DMP-1 Western blotting, PDGF AB and BB treatment groups showed decreased levels of this protein compared with the control group. The expresson in the PDGF AA was higher than that in the control group.

Conclusion: PDGF BB had the strongest proliferative effect on human PDL cells among PDGF AA, AB, and BB. Also, Our results imply that PDGF dimers through DSP and DMP-1 regulate proliferative process on PDL cells.
