The responses of Human pulp cells to PDGF isoforms

Objective:In this study, we investigated the effect of PDGF dimers on the differentiation of pulp cells in vitro through expression of DMP-1.

Methods:Human pulp cell primary culture, calcium content measurement, DMP-1 Western Blot analysis

Results:1. The calcium content in the cell layer in the PDFG AA, AB and BB treatment groups decreased that in the control group; the PDGF AB and BB treatment groups showed lower values than the other two groups. 2. In DMP-1 Western blotting, the PDGF AA treatment group (Figure 2, lane 2) showed increased levels of these proteins compared with the control group. The expresson in the PDGF AB treatment group was similar to that in the PDGF BB group , and the expression in both of these groups was lower than that in the control group.

Conclusion:PDGF AA selectively inhibited the differentiation in the proliferative phase and had no effect on the latter differentiation process subsequent to the early process in the dental pulp cell culture, but PDGF AB and BB had effect on the latter differentiation process by stimulating DMP-1 expression.
