Expression of Extracellular Marix Protein in hGF by Gargling Solutions

Objectives: The purpose of this study is to evaluate the expression of extracelluar matrix protein, such as matrix metalloproteinase -1 (MMP-1), tissue inhibitor of metalloproteinase -1, -2 (TIMP-1,-2), collagen type I, fibronection, in human gingival fibroblasts (hGF) treated with 6 oral gargling solutions, Chlohexin, Heksamedin, Caregargle, Senstime, Garglinmint, and Gijimint. Methods: All experiments were performed using fifth-passages hGF cells. For the MTT and reverse transcription-polymerase chain reaction (RT-PCR) procedures, the fifth-passages hGF cells (2 C 104 cells/well) were treated with 6 oral gargling solutions for during 30 minutes for 1 day or 6 days at 37oC in humidified air with 5% CO2. At the end of the incubation period, the cells were washed with Hank's Balanced Salt Solution (HBSS, Gibco BRL, USA) and processed for MTT and RT-PCR. Untreated cells in Dulbecco's Modified Eagles Medium (DMEM, Gibco BRL, USA) culture medium containing 10% fetal bovine serum (FBS) and saline-treated cells served as controls. The absorbance for cell proliferation was analyzed in 595 nm using Elisa Reader (Bio-TekTM Instruments Inc. USA). For RT-PCR, total RNA was extracted using Trizol Reagent (Invitrogen, USA) and mRNA expression was analyzed by RT-PCR. GAPDH served as the internal control. Results: Cell viability of the hGF treated with 6 oral gargling solutions was decreased than that treated with the control. MMP-1 was strongly expressed in the experimental groups than in the control group, but TIMP (1,2), collagen type I, and fibronectin were expressed in the control group only, by RT-PCR. Conclusion: In these limited experiments, these data suggest that various oral gargling solutions used routinely for periodontal and implant patients can delay the wound healing.
