Methods: We used MPDL6 (murine periodontal ligament cells) which were derived from BALB/c mice. Expression of TLR 1-9 in MPDL6 was examined by RT-PCR. Production of IL-6 and CXCL-10 by MPDL6 after stimulation with TLR ligands was examined by ELISA. Expression of surface marker such as B7 costimulators (B7H1 and CD86) on MPDL6 after stimulation with TLR ligands was examined by flow cytometry. In addition, CD4 T cell proliferation cocultured with MPDL6 was measured by BrdU incorporation and IFNg production from CD4 T cells was determined by ELISA. The effects of coculture of MPDL6 on CD4 T cell activation on the proliferation and production of IFNg.
Results: MPDL6 expressed mRNA for TLR1-7 and produced IL-6 and CXCL-10 after stimulation with TLR ligands. Production of IL-6 and CXCL-10 was suppressed by ROS inhibitors. TLR3 ligand, Poly(I:C) upregulated the expression ofB7-H1 on MPDL6. MPDL6 suppressed proliferation of CD4 T cells and IFNg production.
Conclusions: These results suggest that PDL can act as immune cells and be involved in immunopathogenesis of periodontal diseases.
