Methods: EDBL were prepared with Doxil® and perfluoropropane gas. Intracellular deliveries of doxorubicin were performed by sonoporation with Doxil® or EDBL. Cell viability was determined by staining with trypan blue dye. The cell cycle was analyzed by a flow cytometer. Cells were cultured for 24 h and stained Hoechst 33342 to detect apoptotic cells.
Results: Cell viability was significantly reduced by combination of EDBL and ultrasound treatment. EDBL treated with ultrasound led to an augmentation of the proportion of the cells in the sub G1 phase. A DNA fragmentation was seen at 24 h after EDBL and ultrasound treatment.
Conclusion: The combination of EDBL and ultrasound was found to enhance the cytotoxity of Ca9-22 cells treated by doxorubicin. These findings suggest that combination of EDBL and ultrasound might be an effective drug delivery system against cancer cells.
