HDAC inhibitor induces cell cycle arrest in myeloma cells

Objectives: Histone deacetylase inhibitors (HDACi) are known to achieve to be the expected results as anti-cancer drugs for solid tumors. It is reported that HDACi are presently undergoing clinical trial for treatment of the cancer. In this present study, we evaluated the effect of HDACi to cell cycle in myeloma cells, HS-72, and investigated its molecular mechanism.

Methods: The myeloma cell line HS-72 was treated with HDACi (25-200 ng/ml) for 48 h. Cell viability was determined by WST-1 assay. The cell-cycle was analyzed by a flow cytometer. Western blot analysis was used to detect the expression of cell-cycle regulatory proteins, p21, Rb and CDK4, 6.

Results: HDACi inhibited the cell viability in dose-dependent manner in WST-1 assay. Flow cytometer analysis showed that HDACi induced a G1 phase cell-cycle arrest in HS-72. HDACi-treatment was found to upregulate the expression p21, downregulate CDK6, and induce phosphorylated Rb by Western blotting analysis.

Conclusion: Our results indicated that HDACi induced G1 cell cycle arrest through the regulation of protein related to cell cycle in myeloma cells.
