Effects of melatonin on rat bone marrow cells

Objectives: Melatonin, a lipid-soluble hormone secreted by the pineal gland, is known to regulate the sleep cycle. Recent studies have shown that melatonin is also produced by the retina, brain, spinal cord and gastrointestinal tract and has anti-oxidative and anti-aging actions. The purpose of this study was to investigate the role of melatonin in bone formation and resorption in rat bone marrow cells. Methods: Rat bone marrow cells were incubated at 37°C in aMEM medium supplemented with 0.4% gentamicin and 10% fetal bovine serum. Subconfluent rat bone marrow cells were incubated with and without 50mM of melatonin for 6 and 18 hours to extract RNA. The mRNA levels of alkaline phosphatase (ALPase) and RANKL as indicators of bone formation and bone resorption, respectively, were analyzed with real-time PCR (7500 Fast Real-Time PCR System). Results: ALPase activity increased significantly (p<0.05) and RANKL activity decreased significantly (p<0.05)after 18 hours of incubation with melatonin. Conclusion: These results suggest that melatonin promotes bone formation and suppresses bone resorption in rat bone marrow cells.
