Amelogenin without N-terminal domain affects the metabolism of cementoblasts
Objectives: Amelogenin is a major enamel matrix protein (EMP) produced by ameloblasts. EMP plays a crucial role in periodontal tissue regeneration as well as amelogenesis. It has recently been suggested that amelogenins induce the regeneration of periodontal tissue cells. However, various isoforms of amelogenins derived from full-length amelogenin exert different functions.The purpose of this study was to examine the effects of human amelogenin without N-terminal domain (amg-20) on the proliferation and differentiation of cementoblast lineage cells.
Methods: Amg-20 was synthesised and purified. Cultured human cementoblast-like (HCEM) cells were stimulated by 0-10 µg/ml amg-20.The cell proliferation was evaluated by use of a cell proliferation ELISA BrdU kit and MTS method. Real-time-PCR was performed with light Cycler system using SYBR Green to examine the mRNA levels of alkaline phosphatase (ALP) and osteocalcin (OCN). The amount of calcium was quantified was to examine the effects of amg-20 on differentiation of HCEM cells.
Results: Proliferation of HCEM cells was enhanced significantly by the treatment with amg-20. In addition, mRNA levels of ALP and OCN were up-regulated significantly by the treatment with amg-20 as compared to the control groups. Calcium concentration was also significantly increased by the treatment with amg-20 when compared to the control groups.
Conclusions: It is shown that amg-20 significantly enhances the proliferation and mineralization of cememtoblasts as well as full-length amelogenin. It is thus suggested that amg-20 consists of functional domain of amelogenin in regulating periodontal regeneration.
