Biocompatibility of a new developed material for retrograde fillings

Objectives: The aim of this study was to investigate the biocompatiblity of a new material compared with 2 different materials widely use in retrograde endodontic treatments.
Methods: Mineral Trioxide Aggregate (MTA) and Intermediate Restorative Material (IRM), two widely used materials in endodontic retrograde fillings, and the new suggested material were opposed into popsicle insertes. The toxicity of the materials was tested using tissue cultures of RAW 264.7 macrophage cell lines grown in 96 well plates.. The viability of the cells were measured by XTT cell proliferation assay. Cells subjected to popsticles without any materials served as negative control and ENDOFLASS, a well-known cytotoxic material, served as positive control . Setting time of each material and the toxicity of each material using descending concentrations were measure following overnight culturing.
Results: The new suggested material demonstrated less cytotoxicity compared to IRM, and similar cytotoxicity as MTA - the gold standard material for retrograde endodontic filling. There was no significant influence of setting time on the biocompatibility of the new suggested material compared to IRM and MTA, for three different setting time. Different concentrations of the new suggested material had no influence on cytotoxicity on this invitro model..

Conclusions: The results show that the new material has similar biocompatible characteristics compared to MTA.Setting time and material concentrations had no effect on its biocompatibility. Comparing the new material with the gold standard material- MTA,the results of this in vitro study lay the ground for the next step of in vivo experiments to prove the safety of this new material as a new retrograde filling material.
