Antibacterial and Antiplaque Effects of Some Restorative Materials

Objectives: This study aimed at evaluating the antibacterial and antiplaque effects of some restorative materials on Streptococcus mutans. Methods: Test materials comprised; a resin modified glass ionomer (Vitremer, 3M), a compomer (Compoglass F, Ivoclar), a giomer (Beautifil, Shofu) and a non-fluoride containing composite (Renew, Bisco). Seven disks samples (6mm dia x 2mm thick) of each material were prepared in stainless steel molds and sterilized. Bacterial suspensions of S. mutans NCTC 10499 in Trypticase soy broth with 5% sucrose were grown in sterile tubes containing materials for 48h at 37°C under anaerobic conditions. Bacterial suspensions without material samples were used as control. Changes in optical density (OD) in the test suspensions, OD of bacterial cells adherent to the samples and OD of cells adherent to walls of the tubes were measured at 640nm using a visible spectrophotometer. Antibacterial effect was assessed by percentage inhibition of growth = 1 – OD Test Total/OD Control x 100. Test Total OD = OD test suspension + OD material adherent cells + OD tube adherent cells. Antiplaque effect was assessed indirectly by percentage distribution of cells adherent to material = OD material adherent cells/OD Test Total x 100. Statistical analysis was by one way ANOVA and Post hoc Tukey test (p< 0.05). Results:

Material

% Inhibition (mean + SD)

% Distribution (mean + SD)

Vitremer

23.10 ± 7.61 (a)

4.85 ± 0.81 (A)

Compoglass F

14.63 ± 5.93 (a,b)

4.74 ± 1.12 (A)

Beautifil

7.31 ± 3.21 (b,c)

4.23 ± 0.64 (A)

Renew

4.84 ± 6.99 (c)

7.61 ± 1.74 (B)

Similar letters (a,b,c or A,B) indicate no significance (p> 0.05). Different letters indicate significance (p< 0.05).

Conclusions: Vitremer and Compoglass F showed greater antibacterial activity than Renew but Beautifil did not. However, the three materials showed lesser bacterial adherence, thus better antiplaque effects than Renew.
