Salivary Concentration of MMP2,MMP9 and TIMP2 in Oral Cavity Carcinoma
Objectives: Determine the salivary concentrations of MMP2,MMP9 and TIMP2 in patients with Oral Cavity Squamous Cell Carcinoma (OCSCC) compared to healthy controls Methods: A cross-sectional study was conducted on patients that had confirmed histological diagnosis of OCSCC and saliva samples available, and controls. Saliva collection was performed by mouthwash with sterile saline solution. Salivary levels of MMP2, MMP9 and TIMP2 were quantified using ELISA kits for each analyte (Merck KGaA, Darmstadt, Germany) following the manufacturer's instructions. Descriptive and inferential statistics, including absolute and relative frequencies, and t test or Mann-Whitney test, with a p value <0.005. The study was approved by ethics committee [B.CIEFO-112-2023]. Results: A total of 60 patients and 30 controls were included, 43.43%(n=26) were men and 56.67%(n=34) were women, with a mean age of 66 years (±13). 43.33% of patients were smokers and alcohol consumers. 81.36%(n=48) were in the tongue, 5.08%(n=3) in the maxilla, 5.08%(n=3) in the mandible, 5.08%(n=3) in the gum, and 3.39%(n=2) in the cheek mucosa. All cases were diagnosed as infiltrating keratinizing squamous cell carcinoma (conventional). MMP9 exhibited significantly higher concentrations in the case group (936.47ng/ml,±207.17) compared to controls (13.16 ng/ml,±3.98), with a notable difference between the groups (p<0.005). Similarly, MMP2 levels were markedly elevated in the case group (79.93 ng/ml,±14.98) compared to controls (2.06 ng/ml,±0.18) (p<0.005). No significant difference was found in TIMP2 levels between groups (case: 5.60 ng/ml, ±1.11; control: 2.99 ng/ml, ±0.63). Conclusions: The concentration of MMP9 in saliva was found to be higher in patients with OCSCC compared to controls. Conversely, the saliva concentration of TIMP2 was lower than that of MMP2 and MMP9. Mouthwash with saline solution may prove to be an effective method for identifying proteins in saliva. Further studies are needed to establish correlations between saliva concentrations and tissue expression within the same patients, as well as peripheral blood concentrations.
