The study of rat incisor apical bud cultured in vitro
Objectives: The rat incisor had represented no root development since its special apical bud structure maintained the crown fate and grew continuously. So the study wanted to observe the development of the rat incisor apical bud cultured in vitro. Methods: The apical buds were exposed from 5-dpn rat lower insicors, and cultured with the mandible in vitro for 3d. Then the apical buds were observed with HE stained and immunostained with anti-cytokeratin 14 antibody. Results:The apical bud showed similar structures to naturally growing incisors in the mandible after cultured for 3d in vitro, including inner enamel epithelium (IEE), stratum intermedium, and outer enamel epithelium(OEE). But neither HERS nor root structure was observed. The apical buds were immunopositive for cytokeratin 14. Conclusion: It is conclude that no HERS and no root devolopment of rat incisor apical bud cultured in vitro. The rat incisor apical bud also maintained the crown fate while cultured in vitro.
