Induction of Humoral Responses in Newborn Mice by Anti-caries DNA

Objective:To study the specific immune responses of targeted fusion anti-caries

DNA vaccine pGJA-P/VAX in neonatal BALB/c mice, which encode CTLA-4, human Ig, glucosyltransferase(GTF) and cell surface protein(PAc) of Streptococcus mutans(S. mutans).

Methods:Plasmids DNA pGJA-P/VAX were immunized in neonatal BALB/c mice by intramuscular injection(i.m.) and intranasal(i.n.), with high or low doses. Then Enzyme Linked Immune Sorbent Assay (ELISA) was used to check the specific antibodies IgG in sera.

Results:DNA pGJA-P/VAX immunized by intramuscular injection(i.m.) with two doses both induced specific anti-GTF IgG antibodies in sera, which were significantly higher(P<0.01)than controls. Antibody IgG level has no significant difference (P>0.05) between high and low doses. However, antibody IgG level of DNA pGJA-P/VAX induced by intranasal(i.n.) has no significant difference (P>0.05) to controls, also between high and low doses.

Conclusions:pGJA-P/VAX is an effective immunogen and specific humoral immunity in sera was induced in neonatal BALB/c mice by intramuscular injection after pGJA-P/VAX immunization with high and low doses.
