Relationship Between the Expression of Urease Genes and Nickel Ions
Objectives: To obtain the clone of Streptococcus salivarius 57.Ι urease genes and investigate the relationship between the expression of urease genes in Escherichia coli and nickel ions. Methods: Genomic DNA of S. salivarius was isolated and urease genes were cloned into pGEM-T Easy vector. The obtained plasmid was subjected to nucleotide sequence analysis and transformed into E.coli. The recombinant E.coli was grown in medium with and without nickel ions respectively. The expression of urease genes in E.coli was detected by phenol red assay and pH analysis. Results: The obtained clone was proved to comprise urease genes cluster, ureIABCEFGD, of S. salivarius 57.Ι by sequence analysis. Urease activity of the obtained clone was detected positive in E.coli grown in medium with nickel ions. However, without adding nickel ions, the expression of urease genes could hardly be detected. Conclusions: Expression of urease genes cloned in this study depends on the presence of nickel ions. The current clone can be used in further investigating the mechanism and effect of ureolytic activity of oral bacteria.
