Research about Candida Albicans Antifungal tolerance Persister cells

Objectives: A small number of Candida albicans strains survived under the lethal concentration of antifungal drugs are called “persisters”, the aim of this article is to confirm and isolated the persisters in the growth form of planktonic and biofilms, and research about the biological characteristics of persisters.

Methods: By using 96 well plate and test tube for planktonic Candida albicans ,96 wells plates and glass plates combined with 6 well plates for biofilm strains , all the strains were treated with gradient concentration of antifungal drug (from 0.075 to 128 ug/ml) , we isolated the Candida albicans persisters . Reculture the isolated persisters and then test their drug resistance .

Results: The persisters can be isolated in the growth station of both biofilm and plaktonics , but the emergence frequency of persisters of biofilm station is higher than planktonic, about 100 folds. When the isolated persisters were recultured and formed the new biofilm , the characteristics of drug resistance lost .

Conclusion: Similarly to the bacterial persisters , Candida albicans persisters are phenotypic variants of the wild type that arise in a clonal population of genetically identical cells.
