Objectives:To isolate total RNA of C. albicans by Trizol method, modified Trizol method and ultrasonication method respectively, and then to compare the output, purity and integrity of total RNA. Methods:The total RNA of C. albicans 10231 which has grown to stationary phase were isolated by Trizol method, modified Trizol method and ultrasonication method. The value of A260 and A280 were measured by ultraviolet spectrophotometer and agarose gel electrophoresis was carried out. Then the difference of the output, purity and integrity of total RNA isolated by three methods were compared. Results:The output of the total RNA isolated by three methods was 1.904µg,7.208µg and 5.000µg, and the ratio of A260/A280 was 1.896,1.415 and 1.612 respectively. The result of electrophoresis showed that there appeared three clear bands°ª28s rRNA, 18s rRNA and 5s rRNA respectively when isolating RNA by Modified Trizol method and RNA decomposition happened when isolating RNA by Trizol method and ultrasonication method. Conclusion:The output and the purity of RNA isolated from C. albicans by modified Trizol method were higher than those isolated by Trizol method and ultrasonication method. And RNA isolated by Modified Trizol method was more integrated. Modified Trizol method is better than Trizol method and ultrasonication method.
