Comparative proteome analysis of oral verrucous carcinoma and squmaous carcinoma
Objective:To conduct the proteomic analysis of human oral verrucous carcinoma (OVC) and oral squamous cell carcinoma (OSCC), to compare its expression with their normal oral mucosa , to investigate the difference between OVC and OSCC by using two-dimensional electrophoresis (2-DE) and matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDITOFMS).Methods:The total proteins of OVC and OSCC and each patients' normal oral mucosa (OVCN and OSCCN)were separated with 2-DE by using immobilized pH gradient strips and visualized by staining with silver nitrate.The gel images were acquired by scanner and 2-DE analysis software, Image Master 2D Elite. Twenty distinct protein spots were excised from gel randomly and digested in gel by TPCK-trypsin. Mass analysis of the tryptic digest peptides mixture was performed by using MALDI-TOF-MS. Peptide mass fingerprints (PMFs) obtained by the MALDI-TOF analysis were used to search NCBI, SWISS-PROT and MSDB databases.Results: The 2-DE gels showed that protein spots average were 1186±74 in OVC;1076±56 in SCC ;1024±47 in OVCN and 1027±49 in SCCN.There was a good reproducibility of spot position with average 1.562±0.132 mm deviation in pI direction and 1.275±0.232 mm deviation in Mr direction. There were 74 differential protein spots in OVC and SCC, 36 differential protein spots in OVC and OVCN, 31 differential protein spots in SCC and SCCN. Through identification of protein spots from 2-DE gels, 20 PMFs maps were obtained by MALDI-TOF-MS and it were preliminarily identifyied . Conclusion:The methods of analysis and identification of protein spots of OVC and OSCC comparing with each patients' normal oral mucosa in 2-DE gel with silver staining by MALDI-TOF MS derived PMF have been established. Protein expression profile of OVC and OSCC has been obtained. It is demonstrated that OVC is different from OSCC in a change of proteomics and the difference proteins may play a key role in their carcinogenesis.
