Effects of LOH, MSI on 9p, 3p in OLK, EK

Objective: To detect the LOH and MSI of microsatellite loci at chromosome 9p, 3p and their relationship with pathological grades and cell proliferation in OLK, EK and OSCC. Methods: 83 samples were collected, including 39 cases of OLK, 12 cases of EK,32 cases of OSCC. A pair of genetic DNA was isolated from resected tissues and peripheral blood was taken from each patient. Polymerase chain reaction, Polyacrylamide urea gel electrophoresis and silver staining were used to detect 7 microsatellite loci°ªD9S171, D9S1752, D9S1748, IFNA, D3S1266, D3S643 and D3S966. The relationship of LOH, MSI with pathological grades and cell proliferation was studied. Results: Significant differences were found between total LOH in chromosome 9p and/or 3p with pathological grades (P<0.05), so as LOH and MSI and pathological grades. LOH has positive correlation with AgNOR number,so as MSI with PCNA index. The clinical therapeutic effect of experimental group is better than that of the control group. Conclusion: There may be some tumor suppressor genes on chromosome 9p and 3p that were related to the progression of OSCC. LOH and MSI may play a certain role in the process of oral carcinogenesis.
