The effect of lactoperoxidase system on porphyromonas gingivalis trypsin-like protein

Abstract

Porphyromonas gingivalis can transmit between family members through saliva and lactoperoxidase-I--H2O2 system with physiological concentration of SCN- has a strong antibacterial effect on P. gingivalis, but its effect on pathogenesis of P. gingivalis have never been reported yet. Objectives: to investigate the effect of LP-I- -SCN- - H2O2 system on P. gingivalis level of Trypsin-like protein (TLP). Methods:  Pg (ATCC 33277) were grown in Brain Heart Infusion agar plate and dilute to 8.0°Á109/ml, four groups were divided according to the incubation time with lactoperoxidase -I- - SCN- -H2O2: the 0min (10min, 15min, 25min) groups. After incubating on a shaking water bath at 37oC for 0min (before adding H2O2), 10min, 15min and 25min respectively, add 5µl DTT to abolish the oxidation. Then examine the level of TLP through specific kit after cultivated in facultative anaerobic condition for 48h. Results: the overall level of TLP dropped with the growing of cultivation time: It dropped significantly from 147.679 (u/ml) to 83.818 and 38.756 when cultivated with  LP-I-- SCN- -H2O2 system for 15min and 25min respectively (P<0.05). Conclusions: The LP-I--H2O2 system with physiological concentration of SCN- can effectively decrease the level of TLP of P. gingivalis. This result will provide with substantial evidence for further developing oral health products concerning anti-periodontal diseases with lactoperoxidase-SCN- -I- - SCN- -H2O2 system.
